Changes in Atlantic cod (Gadus morhua L.) sperm quality during the spawning season ArchiMer
Rouxel, Catherine; Suquet, Marc; Cosson, J; Severe, Armelle; Quemener, Loic; Fauvel, Christian.
The biology of cod reproduction is well described in the scientific literature. However, sperm biology and spermatozoa management are poorly studied in this species. Because of its recent farming expansion, a better knowledge of cod gametes is becoming especially useful. This work aimed at establishing tools to study sperm biology in cod, and also investigated the existence of changes in cod sperm quality during the spawning period. We showed that sperm concentration could be assessed using spectrophotometry at 260 nm. Sperm motility significantly decreased after a 168-h storage at 4 degrees C. A 1:9 dilution of sperm in a non-activating medium (1/3 seawater and 2/3 freshwater, osmotic pressure: 360 mOsm kg(-1)) improved sperm storage. Sperm concentration,...

Characterization of sperm motility in sea bass: the effect of heavy metals and physicochemical variables on sperm motility ArchiMer
Abascal, F; Cosson, J; Fauvel, Christian.
Computer assisted sperm analysis (CASA) was used to characterize the motility of sea bass Dicentrarchus labrax spermatozoa and to study the effect of several physicochemical variables and heavy metals on sperm swimming performance. Duration of sperm motility in sea bass was very short (< 50 s). During the first 20 s all the motility variables measured remained approximately constant, the velocity and linearity of the movement being maximum during this period, while both variables decreased sharply later. While slight variations in pH did not significantly modify sperm swimming performance, changes in osmolality affected all the measured motility variables. Two of the heavy metals tested, Cu2+ and Pb2+, did not affect sperm motility when the activating...

Cryopreservation of sperm in marine fish ArchiMer
Suquet, Marc; Dreanno, Catherine; Fauvel, Christian; Cosson, J; Billard, R.
Since the first work of Blaxter in 1953, fish sperm cryopreservation has been attempted on about 30 marine species. The present paper reviews the techniques used and the results published in these species. Particular attention is paid to the handling procedure of sperm before freezing, the problems of semen ageing and semen contamination with urine. The quality of frozen-thawed semen was evaluated using previously standardized biotests, such as a two-step motility activation technique adapted for the different species and fertilization assays using a discriminating insemination technique. Most extenders used in marine fish are saline or sugar solutions. From the investigated cryoprotectants, dimethyl sulphoxide (DMSO) generally leads to the best results....

Experimental processes of hybridization in the Laminariales of French coasts. ArchiMer
Cosson, J; Gayral, P.
The research has shown that interspecific and intergeneric hybridization is possible in the Laminariales with variable rates according to the species used; these rates were determined precisely only following preliminary study where the rate of parthenogenetic development was evaluated for each particular species. Besides, the results obtained concerning the ecophysiology of the Laminariales and especially the effect of temperature and light on the fertility of the gametophytes, prompt to undertake experimental cultures of the hybrids in order to obtain algae with qualities superior to those of the parents, particularly regarding alginic acid content and growth. The experiments carried out in 1983 concern together the rearing ashore of isolated...

Marine fish spermatozoa: racing ephemeral swimmers ArchiMer
Cosson, J; Groison, A; Suquet, Marc; Fauvel, Christian; Dreanno, Catherine; Billard, R.
After along period of spermatogenesis (several weeks to months), marine fish spermatozoa are delivered at male spawning in seawater (SW) at the same time as ova. In some fish species, as the ova micropyle closes quickly after release, these minute unicells, the spermatozoa, have to accomplish their task of reaching the micropyle within a very brief period (several seconds to minutes), for delivery of the haploid male genetic information to the ova. To achieve this goal, their high-performance motile equipment, the flagellum, must fully activate immediately on contact with the SW and then propel the sperm cell at an unusually high initial velocity. The cost of such 'hyperactivity' is a very rapid consumption of intracellular ATP that outstrips the supply....

The quality of Pecten maximus sperm ArchiMer
Faure, Catherine; Devauchelle, Nicole; Girard, J.p; Cosson, J.
Sperm of the scallop Pecten maximus (L.) can be collected by various methods for artificial reproduction. Characteristics such as spermiation time, fertilizing capacity, respiration and motility of the spe can be modified by collection method. In the current study, sperm was collected quickly by scarification, but its quality was only average. When thermal shock or serotonin injection was used to stimulate the release of sperm, it was collected and concentrated as "dry" as possible after exit fr the kidney. Its quality was comparable to that of diluted sperm collected after thermal shock, a technique commonly used in bivalve hatcheries. The results also showed that serotonin injection systematically decreased spermiation time and induced the production of...
